A Standard Manufacturing Method

Solid-phase peptide synthesis, commonly abbreviated as SPPS, is the standard technique used to manufacture peptides for research purposes. Developed decades ago, it remains the dominant method because of its reliability and scalability.

Building the Chain One Amino Acid at a Time

In SPPS, the peptide chain is built one amino acid at a time on a solid resin support. Each amino acid is added in a stepwise cycle involving coupling, washing, and deprotection, allowing the chain to grow in a controlled, sequential manner.

Why a Solid Support Helps

Anchoring the growing peptide chain to an insoluble resin bead simplifies purification between synthesis steps. Excess reagents and byproducts can simply be washed away, since the peptide itself remains attached to the solid support throughout the process.

Cleavage and Purification

Once the full sequence has been assembled, the completed peptide is cleaved from the resin and undergoes purification, typically using high-performance liquid chromatography, to separate the target peptide from any incomplete or side-reaction products.

Quality Depends on Process Control

Because each coupling step needs to go to completion for a pure final product, tight process control at every stage of synthesis has a direct impact on the purity and consistency of the finished peptide, which is why manufacturing standards matter so much in this field.
