What Reconstitution Means

Reconstitution is the process of returning a lyophilized, freeze-dried compound back into a liquid solution, typically by adding a sterile diluent. This is a standard step in many laboratory workflows involving peptides and other biomolecules.

Choosing an Appropriate Diluent

The diluent used during reconstitution needs to be compatible with the compound being dissolved. In laboratory settings, sterile water or buffered solutions are commonly used, chosen based on the specific properties of the compound and the requirements of the intended experiment.

Technique Matters

Adding diluent slowly along the side of the vial, rather than directly onto the lyophilized powder, helps avoid excessive foaming or agitation that could affect the compound’s structure. Gentle swirling, rather than vigorous shaking, is generally recommended to fully dissolve the material.

Maintaining Sterility

Because reconstituted solutions are more susceptible to microbial contamination than lyophilized powder, working under sterile conditions and using proper aseptic technique is an important part of any reconstitution procedure in a research setting.

Stability After Reconstitution

Once reconstituted, most compounds have a shorter stable shelf life than in their lyophilized form and are generally more sensitive to temperature. Documenting the reconstitution date and following appropriate storage conditions afterward is good laboratory practice.
