How HPLC Measures Purity

HPLC separates the components of a sample as they pass through a column, based on differences in how strongly each component interacts with the column material. The result is a chromatogram showing distinct peaks, where the size of the main peak relative to the total area indicates the sample’s purity percentage.

How Mass Spectrometry Confirms Identity

Mass spectrometry measures the mass-to-charge ratio of ionized molecules in a sample, which allows researchers to confirm that a peptide’s molecular weight matches its expected sequence. This is essential for verifying that the correct peptide was actually synthesized.

Why Both Are Needed

HPLC alone can show a clean, single peak without confirming what that peak actually is. Mass spectrometry alone can confirm identity without necessarily reflecting overall purity. Used together, the two methods cross-check each other, giving a much stronger basis for quality verification.

What This Means for Researchers

When evaluating a research peptide supplier, checking whether both HPLC and mass spectrometry data are included in a certificate of analysis is a useful way to gauge how seriously that supplier approaches quality verification.
